Deconstructing translation with your own cell line

Studying translation in vitro can provide unprecedented resolution into a cellular system, allowing you to assess how cellular stress affects translation, accelerate mRNA drug screening, among others. Immagina’s in vitro translation services reconstitute a cell-free cellular system – totally customized. Starting with the cell line of your choice, we isolate ribosomes, cytoplasmic extract and combine them with a reporter mRNA. After the client’s approval, we produce these components, making it easy for you to test the effect of different translation components on your desk.

Service overview

Leveraging the work of Dr. Anders Lund at the Biotech Research and Innovation Centre, University of Copenhagen, Denmark1, our service takes it one step further, yielding higher and more stable protein signals than the standard protocol. We provide a full end-to-end solution:

  1. Cell type on-boarding & growth: customized to your needs. K562 and HEK-293 - cell lines optimized and available in house. Other cell lines available on request:
    • CHO
    • HeLa
    • U2OS
    • A549
    • Jurkat/THP1
    • Any other cell line (externally provided)
  2. Translation components preparation: after cell growth, lysis and ultracentrifugation are performed to obtain isolated ribosomes and cytoplasmic extract.
  3. Validation of translational activity: confirmed via luciferase reporter mRNA and further tested with dose-dependent translational inhibitors
  4. Execution: upon validation of the in vitro translation system, we proceed with production of the cell line. After isolation of ribosomes and cytoplasmic extract, we start producing your customized kit.
  5. Product delivery: You will soon receive a validated kit, ready to use on your bench with a standardized protocol.
