We developed RiboLace, a new method for the isolation of active ribosomes by means of an antibody-free and tag-free pull-down approach based on a new puromycin-containing molecule. RiboLace is fast, requires very low input material and can be rapidly used to obtain a global snapshot on the active ribosome footprints at single nucleotide resolution.
No other tolls can generate a snapshot of protein expression with such degree of resolution and accuracy.
Why a better ribosome profiling?
- Thanks to a novel method based on an original puromycin-containing molecule (patented) capable of isolating active ribosomes.
- It can be easily and rapidly applied both in vitro and in vivo, thereby generating a global snapshot of active ribosome footprints at single nucleotide resolution.
- Fast protocol (10x). Not more 24 hours of polysome profile required.
- It works reliably with low amount of input material. Less than 10^5 cells are required for isolation of active ribosome protected fragments.
